Unique gut microbiome patterns linked to healthy aging, increased longevity

Biological changes in the gut microbiome as we age may be connected to overall healthy aging and increased survival rates, according to NIA-supported research published in Nature Metabolism. Learning more about the balance of this microbiome — all the microorganisms, bacteria, viruses, protozoa, and fungi, and their collective genetic material in the gastrointestinal (GI) tract — is an evolving scientific field. Current findings suggest potential connections to many health conditions including obesity, metabolic disorders, inflammation, cancer, and depression.

Throughout the human life span, gut microbiome follows some predictable patterns, with rapid change from infancy to age three, stability up until middle age, and then accelerated change starting in late adulthood. Similar to fingerprints, no two microbiomes are exactly alike but there is a common group of microbe types found in everyone, comprising about 30 percent of the total.

Previous studies had found gut microbiome pattern differences between older adults who are lean and physically active compared to their less fit and healthy contemporaries. Other research connected early development of frailty to less gut microbial diversity.

For this newer study, the research team analyzed gut microbiome genetic sequences and a wealth of other health and survival outcomes data from more than 9,000 people between the ages of 18 and 101. They paid particular attention to a longitudinal study of a community of over 900 older adults to get a clearer picture of the makeup of the mature GI microbiome.

The investigators found that older adults who had a more unique pattern of changes to their GI microbe profile also tended to be healthier and live longer than peers with less microbiome divergence. The healthier participants’ blood tests showed lower levels of LDL cholesterol and higher levels of vitamin D, but also more beneficial blood metabolites produced by gut microbes. One of these metabolites, tryptophan-derived-indole, has been shown to reduce inflammation and to extend lifespan in mouse studies.

People whose gut microbiomes had grown more unique with age were able to walk faster and had better overall mobility than peers who showed less GI microbe changes with age. Plus, those with less diverse gut environments used more medications and were nearly twice as likely to die during the study period.

The research team suspects that modern diets rich in salty, sugary, or fatty processed foods may damage the aging gut, while higher fiber nutritious foods like fruits, vegetables, seeds, beans, and nuts could be combined with exercise to help protect a healthy gut microbiome balance as we age. More studies will enable continued explorations into the links between our body’s gut microbes and overall healthy aging, especially the role of diet and physical activity.

Dragon Age teaser impressions and reflections on the future of the series

At today’s The Game Awards, we were shown a mysterious teaser for the new part of Dragon Age and. .. that’s it. Promising hype and allusions on social media turned out to be empty shells. Let’s talk about what we have and outline the problems that await BioWare if they want to return to the big leagues.

Old Guard

Varric was entrusted with a new story to tell. A dwarf with a charming voice and a rich imagination pulled the blanket over himself for the entire second part, and in The Inquisition he lost the lion’s share of charm. It seems that BioWare realized that they couldn’t come up with a better character — and returned one of the main roles to him.

Of course, Solas, the Dire Wolf and the secret villain from the Inquisition, will return. We broke up with him when he freed the Inquisitor from the mark (albeit with his hand) and threatened to destroy the world. I hope that in the new part of the story, Solas will really try to erase Thedas into dust: even the most devoted fans cannot stand another cardboard villain like Corypheus.

Did you miss me?

Break — do not build

The title (yet?) has no number or clarification. Dragon Age is Dragon Age. Does this mean a reimagining of the series? Dont clear. But the developers’ flirting with the cyberpunk style inspires much more fear: look at the picture below.

Jumping into a hype engine is not easy. But why do it at all?

I really hope that BioWare will not stuff the game to the eyeballs with fashionable ideas. «Pointless but cool» — is not about Dragon Age at all. The trailer flashed a flock of giant turtles on the beach, and a snowy forest, and an almost fabulous Tevinter castle. There is no doubt that our eyes will again run out from visual changes. But another problem arises: will all these locations be needed by ? Remember the endless voids of the Inquisition, where half of the locations could be driven on a mount without stopping. Of course, you should not get to the forced crampedness of Dragon Age II, where we spent the entire game literally in the same locations, but please, let the locations be at least beautiful in the new game.

The appearance of enemies, environments and characters is pointless to discuss yet, but here BioWare has never been wrong. I sincerely hope that in this part the developers will finally get their hands on the animation of faces — in the «Inquisition» she literally destroyed some of the dialogues with her unnaturalness.

I couldn’t understand anything else from the video: in fact, I can’t even call it a trailer for a Dragon Age game. The recognizable frescoes and voice of Varric, as well as the qunari, and the darkspawn, and the mysterious Solas, do not add up to a single picture. Someone walks / runs / sits / fights, but do the developers really have until now (and the first teaser came out exactly two years ago) no central idea worth showing?

Heavy Legacy

Today’s BioWare, which has little in common with the studio that made the first games, got a difficult franchise. This is what the Dragon Age series looks like in the eyes of most: epochal Origins, crumpled second part and Inquisition — at the same time the crown of the evolution of the series and a rare hat. This fall, I replayed all parts of the series and I can say for sure that each part is imperfect, but it’s a miracle how good it is in certain aspects. Where the first part took its toll with honest variability, the second part was furtively credible with small stories of Hawke’s environment, and the third part was a kaleidoscope of living characters.

The path of Dragon Age is not like evolution: the series rushed about within the RPG genre. Games borrowed elements of CRPG, ARPG, mmorpg, but the proportions changed each time. During the autumn marathon, I often had the feeling that these are three games that are completely different in spirit. I think the mood swings of the games only exacerbated the strife between fans of the series. Many preferred to take the position “before it was better”, the neophytes settled in the “Inquisition” (not forgetting, however, to set mods to reset the command rate timer), and rare apostates glorify the second part. Gluing together such different audiences (and attracting a new one!) is a monstrously difficult task. Probably BioWare will try to sit on all the chairs at once — and from some0091 will inevitably fall .

Is there an ace up your sleeve?

The unshakable pillar and cementing foundation of the Dragon Age series remains the game world. The simultaneous flow of information through the mouths of the characters, hundreds of cultural items and tons of notes help players to put together a coherent picture of Thedas. At the same time, sources often contradict each other (for example, elven legends, dwarven Chronicles and the Song of Light describe the creation of all things in different ways), which only encourages players to explore the world, ask questions and find answers.

However, there are serious concerns about the departure of several key employees from BioWare. Mike Laidlaw and David Gaider, who had a hand in all the parts, left the studio more than two years ago, and last week BioWare head Casey Hudson himself and Dragon Age 4 executive producer Mark Darra packed the suitcase. Will the remaining members of the team be able to cope with the franchise, preserve and develop the lore of Thedas? Only time and gameplay will show — neither from trailers, nor from developer diaries, we will understand this.

«And then Flemeth came and saved everyone» is an integral plot twist of every Dragon Age. At the same time, he surprisingly logically correlates with the rest of the storylines and actually builds his own: for example, the scene with the rebirth of Flemeth from the medallion at the beginning of the second part takes on a new meaning when we find out who she really is. Judging by the epilogue of the third part, Flemeth will have to look for a replacement. Someone should definitely say a cryptic phrase like “I push the story when I need to with a grin. But sometimes you have to kick it.»

A good start is half done

In the trailer, we were pathetically informed that the time has come for a new hero, «without a magic hand and an ancient prophecy. » It’s funny that in the first and second parts there were no prophecies and a sense of «chosenness» — the heroes were just trying to swim out of the cycle of events. Has BioWare forgotten who they make games about?

That’s why the prologue is my biggest concern. He managed the studio worse and worse with each subsequent part. The bar for Origins will probably not be reached: 6 prologues are hardly needed, each of which affects the plot in its own way and reveals one of the parts of Thedas. But it is impossible to break through the bottom deeper than the beginning of the Inquisition. The set of clichés «You are the survivor-the-chosen-leader with no backstory» and a few hours of grinding in the Hinterlands caused many to abandon the game. It will not be a shame to repeat the beginning of the second part: Varric tells the story of a new hero, generously seasoning the truth with fiction.

There will be a fight

The combat system in the series has changed the most. Playing after Inquisition in the first part is very strange: viscous battles, slow swings and the death of the party due to an undercontrolled genlock emissary who managed to throw Crushing Dungeon at the robber. It is unlikely that the mass audience will like such a fight in jelly, as well as the jerky fights of the second part.

BioWare must remember about the dying section «Tactics». Next to the first part, where you can incredibly flexibly customize the behavior of a party member or take a ready-made set, Inquisition looks like a clumsy fake. Half of the third part of the soldiers in my detachment were not beaten simply because they forgot how it was done. I had to manually give the command to deliver a normal blow. So mocking the players is unacceptable: you need either the revival of Tactics, or the painstaking work of BioWare themselves on artificial intelligence.

Magic should serve man, not man of magic

As an inveterate sorcerer in all games, I demand the return of the mage from Origins. What is called a «magician» in the third part — 8 buttons, one of which will definitely be a barrier, and another one will be the ability to concentrate, which is pressed only on big holidays — is not a magician. I don’t want to press the normal attack button 3/4 of the time anymore.

I perfectly understand the annoyance of all the «tanks» getting stuck in the runes of paralysis, the Storm of the Century and other entropies. I played the rogue myself and hated the deadly sairabaz. But mages in Dragon Age have always been portrayed as an unbridled force that is sometimes easier to destroy than to try to control.

They decided to balance them very simply: they destroyed the variability. 68 + 8 spells in the first part, 32 + 8 in the second, during the “Inquisition” turned into worthless … 18 + 4 from specialization. Healers, controllers and buffers, dear to the heart, but optional to the highest levels of difficulty, were declassed, instead of which there was a “seven-bads-one-answer” barrier, without which it is almost impossible to play already on medium difficulty. And give us blood magic back! I really want to NOT use these charms.

Mandatory additions

The hallmark of the series has become DLC, which script and gameplay wipe the nose of the main parts. «Awakening» accelerated the phlegmatic Origins and introduced even more variability, other additions helped to assemble the mosaic of the world. Without the Killer Brand to rip Hawk out of the cold Kirkwall, without the Legacy to introduce us to the antagonist of the third part, Dragon Age II feels different. And the fact that the true ending of the «Inquisition» and unconditionally the best plot twist of the series ended up in the «Outsider» expansion comes to the point of absurdity. All in the same third part, the most important aspects of the lore (the culture of the mysterious Avvars, the mystery of the first Inquisitor and the origin of the lyrium) and the most atmospheric locations ended up in the DLC. It seems pointless to complain about this: I expect that the situation will repeat itself in the new part. The complete edition is strictly required.

Titans, ancient elves and the mysteries of the Gray Wardens — The Inquisition has cast enough hooks to continue. However, one of the most powerful characters in the series did not receive development: the intelligent creature of darkness, the Architect, did not appear in the following parts of the series. His good intentions (freeing the darkspawn from the grip of the fel) and their consequences (bloodbaths and raising crazed monster incubators) might make him the most controversial villain in the series. The most daring plot I’m guessing is that the Architect will offer aid to the Inquisition and a darkspawn horde (or a new Archdemon!) will come to Thedas’ defense. And where the Architect is, there is his elf companion Seranni and her sister Velanna from the same Awakening. And certainly the case will not do without the elves from the second part: Merrill and Fenris.
